High-performance liquid chromatography — the standard method for measuring how pure a peptide sample is.

In plain terms

HPLC separates a sample into its components and measures the relative size of each. The result is expressed as area purity: the percentage of the detected material that is the target peptide.

It measures purity relative to what the detector can see. Salts, water, and bulking agents are invisible to it, which is why HPLC purity and net peptide content are different numbers.
